Special Topic Introduction

Cardiovascular aging represents a central biological process underlying the increasing prevalence of cardiovascular diseases worldwide. Increasing evidence indicates that age-related cardiovascular dysfunction arises from the interplay between genetic predisposition and complex epigenetic and genomic regulatory mechanisms that progressively reshape cellular identity and tissue homeostasis.

Recent advances in epigenomics and three-dimensional (3D) genome technologies have revealed that chromatin organization is a key layer of regulation for cardiovascular gene expression. Spatial genome regulatory mechanisms orchestrate coordinated transcriptional programs that govern vascular homeostasis, cardiac remodeling, as well as inflammatory and stress responses, while their dysregulation contributes to progressive cardiovascular decline. In parallel, integrative multi-omics approaches are enabling unprecedented resolution of cell-state-specific regulatory landscapes during cardiovascular aging and disease progression.

This Special Issue aims to provide an integrated platform for understanding the mechanisms underlying cardiovascular aging and disease progression. It addresses how genetic and epigenetic regulation, together with cellular and molecular interactions, shape cardiovascular function in aging across basic, translational, and clinical contexts. We welcome studies that offer mechanistic and translational insights to advance therapeutic strategies for age-related cardiovascular diseases.
