Special Topic Introduction
Cancer is a highly heterogeneous and evolving disease driven by continuous genomic alterations and evolutionary processes. Advances in next-generation sequencing and computational genomics have revealed that tumor initiation, progression, and treatment outcomes are profoundly influenced by somatic mutations, genomic instability, clonal evolution, and functional changes within cancer genomes. Understanding the dynamic evolution of tumor genomes is therefore essential for uncovering disease mechanisms and advancing precision oncology.
Recent studies have demonstrated that driver mutations, structural variations, DNA repair defects, and other genomic alterations shape tumor evolution and influence clinical responses. Meanwhile, emerging functional genomics approaches and integrative computational strategies are enabling the discovery of cancer vulnerabilities, molecular biomarkers, and new therapeutic opportunities. Deciphering how tumor genomes evolve and adapt under different selective pressures will provide critical insights for developing more effective and individualized treatment strategies.
This Special Issue, entitled “Tumor Genome Evolution and Precision Oncology: From Genomic Alterations to Therapeutic Adaptation,” aims to highlight recent advances in cancer genomics and its translation into precision medicine. We welcome original research articles, reviews, and perspectives focusing on the mechanisms of tumor genome evolution, functional consequences of genomic alterations, and emerging genomic approaches for improving cancer diagnosis, prognosis, and treatment.
Topics include, but are not limited to:
● Tumor Genome Evolution and Clonal Dynamics
Studies exploring the evolutionary processes of cancer genomes, intratumoral heterogeneity, clonal selection, and genomic trajectories associated with tumor development and progression.
● Driver Mutations and Functional Consequences of Cancer Genomic Alterations
Research focusing on the identification, characterization, and functional validation of driver mutations, oncogenic alterations, and their roles in cancer initiation and progression.
● Genomic Instability and DNA Repair Mechanisms in Cancer
Studies investigating genomic instability, DNA damage response pathways, mutational signatures, and their contributions to cancer evolution and therapeutic opportunities.
● Functional Genomics and Discovery of Cancer Vulnerabilities
Research utilizing functional genomics approaches, including CRISPR-based screening, genetic perturbation technologies, and integrative omics analyses to identify novel cancer dependencies and therapeutic targets.
● Genomic Determinants of Treatment Response and Cancer Adaptation
Studies examining how genomic alterations and evolutionary processes influence tumor responses, adaptive changes, and clinical outcomes following targeted therapies, immunotherapies, and other treatment strategies.
● Cancer Genomic Biomarkers and Precision Oncology Applications
Research focused on the discovery and validation of genomic biomarkers, molecular signatures, liquid biopsy approaches, and genomic-guided strategies for personalized cancer management.
● Computational Genomics and Artificial Intelligence in Cancer Precision Medicine
Studies applying bioinformatics, artificial intelligence, and machine learning approaches to analyze cancer genomic data, predict treatment responses, and facilitate precision therapeutic decision-making.
Through this Special Issue, we aim to bring together researchers and clinicians in cancer genomics, molecular oncology, computational biology, and translational medicine to advance our understanding of how genomic alterations drive tumor evolution and shape therapeutic outcomes. This collection will provide new perspectives on leveraging cancer genomic knowledge to develop innovative precision oncology strategies
