Special Issue Introduction

The tumor microenvironment (TME) is a complex and dynamic ecosystem surrounding cancer cells, consisting of diverse cellular and non-cellular components. Cellular elements include cancer-associated fibroblasts (CAFs), immune cells, endothelial cells, etc. Non-cellular components encompass soluble factors like growth factors and cytokines, extracellular matrix (ECM), and physical properties such as hypoxia.

The intricate network of interactions between tumor cells and the TME plays a pivotal role in tumor progression and metastasis. The TME influences each stage of the metastatic cascade through various mechanisms. For instance, CAFs can promote tumor invasion and metastasis by remodeling the ECM and secreting growth factors that stimulate cancer cell proliferation and migration. Immune cells within the TME can exhibit both anti- and pro-tumor functions. Additionally, the abnormal hypoxic conditions and tumor vasculature in the TME can facilitate cancer cell intravasation and dissemination.

Understanding the intricate interplay between tumor cells and the TME is essential for unraveling the mechanisms driving cancer metastasis and for developing effective therapeutic strategies. Targeting components of the TME that promote metastasis holds promise for preventing or inhibiting the spread of cancer and improving patient outcomes.
